Earlier this month, the ESA finally began making some of its plans for E3 2021 official, announcing press conferences and appearances from a number of large publishers. At the time, Square Enix wasn't included in the list of participating publishers but as it turns out, they will be joining others in making announcements.
E3 2021 is set to take place between the 12th and 15th of June, featuring appearances from Nintendo, Xbox, Ubisoft, Take-Two, Warner Bros, Capcom, Konami, Koch Media and now, Square Enix as well. In an interview with Nikkei XTrend, Square Enix President, Yosuke Matsuda, confirmed that the publisher is “planning to announce at E3 in June”.
We still don't have specific dates for all the press conferences at E3 2021. Ubisoft has been the only publisher so far to announce an exact date and time – you can find all of that information, HERE.
In the meantime, we can speculate on what may appear at Square Enix's portion of the show. Recently, the publisher has announced two major games – Final Fantasy 16 and Life is Strange: True Colours. It seems reasonable to expect that we'll see more of these games at E3, in addition to some updates on smaller projects, like Neo: The World Ends With You, Final Fantasy 7 Remake Intergrade, Nier: Replicant, Project Triangle Strategy and perhaps better look at Marvel's Avengers Black Panther expansion. Hopefully we'll also see some brand new surprises as well.

KitGuru Says: Square Enix usually does well with its E3 shows, so it will be great to see them return again this year. June is stacking up to be a great month for gamers.
